Design, synthesis and screening of cleavable nucleotides as reversible terminators for DNA sequencing by synthesis

We have designed and synthesized a nucleotide library of chemically cleavable and photocleavable nucleotides as reversible terminators for SBS. The library was screened for its ability as a reversible terminator for SBS in the solution phase and the chip system. All the photocleavable nucleotides have been shown to be excellent substrates for DNA polymerase, and photocleavage is rapidly and efficiently achieved by using a UV laser. These four fluorescent-labelled nucleotide terminators (A, C, T, G) with a 3'-O photocleavable group will finally provide a new platform for DNA sequencing in genomics industry.
