Determination of 7,8-dihydroxy-coumrin in roots, stems, leaves, fruit of Daphne koreana Nakai by HPLC

An improved method to determine content of daphnetin (7,8-dihydroxy-coumarin) in roots, stems, leaves and fruit of Daphne Koreana by HPLC. Daphnetin was extracted from powders of the four parts in methanol, and and further extracted using ethyl ether to prepare samples. The determination of daphnetin was performed on ODS-C18 column, the mobile phase was acetonitrile-0.4% of phosphate buffer (40:60, volume/volume), the flow rate was 1.2ml/min, and the detecting wavelength was 327nm at 20℃. Content of daphnetin in roots, stems, leaves and fruit demonstrated a great difference, and the content in leaves is the highest. There is an obvious difference of content among the four parts. The determination of the content of daphnetin in Daphne koreana gives a generally reference for its quality control, also makes a guidance for daphnetin extraction manufacture using Daphne koreana as a natural resource.
