Preparation of 15-N and 13-C enriched heparan sulfate oligosaccharide precursors

Heparan Sulfate Proteoglycans orchestrate various biological functions through specific binding to specific proteins. To define the chemical basis of such biological interactions using NMR methods, we have produced 15N and/or 13C isotope enriched Heparan Sulfate precursor polysaccharides from E.coli K5 strain. The resulting isotopically enriched polymers were cleaved with recombinant heparin lyases to obtain oligosaccharides of various sizes. These oligosaccharide precursors are then subjected to enzymatic modifications to obtain bioactive Heparan Sulfate Oligosaccharides to define Heparan Sulfate-Protein interactions.
