Littorachalcone, a biologically active polyphenol, was found to be useful in the treatment of various neurological disorders including Parkinson's and Alzheimer's disease. A direct and convergent synthesis of littorachalcone will be presented. A bis-aldol reaction was used as a key step involving 2,4-dimethoxyacetophenone and suitably functionalized 4,4'-oxybisbenzaldehyde. Reduction of resulting bis-enone by NiCl2-NaBH4 followed by removal of protecting groups furnished the target molecule in an efficient manner.
