Study on the extraction and separation of AECI from enzymatic hydrolysate of oyster

The oyster shell was a common sort of marine mussels, and its meat and shell serve as traditional medicines. To invest the method in which ACEI ( Angiotensin-Converting Enzyme Inhibitor) can be prepared , this experiment took oyster as study object, employed inhibitory coefficient as standard and utilized tryptase as an enzyme. The optimum technology conditions (E/S 390U/g, 45°C, pH8.5, hydrolysis time 3h , the solid to liquid ratio of 1:2) were obtained according to the result of orthogonal test and orthogonal variance analysis. Moreover, with SephadexG-25 the study carried out the chromatogram separation of the hydrolysate, and also measured inhibition coefficient and relative molecular weight distribution of ACEI from hydrolysate. The result showed that after the chromatogram separation, ACEI determined to be at the highest activity at the second elution peak at which the active coefficient of ACEI was 89%, and that the relative molecular weight distributed between 1842 and 1396Da.
