The folding up of RNA into secondary structures covers the primary structure so that it is no longer accessible and the oligonucleoside targeting these areas has to have a different design around e.g. bulges, and hairpins. In the aim of targeting secondary nucleic acid structures, we have synthesised conformationally restricted nucleosides using different metathesis reactions, both on the monomer and the dimer levels. The conformationally restricted units are believed to bend the oligonucletides so that optimal binding to an RNA containing secondary structures are achieved.
