| Cardiovascular disease is closely related with high cholesterol level, therefore, inactivation of enzymes involved in mevalonate pathway of cholesterol biosynthesis is one major method for treating cardiovascular disease. The mevalonate pathway has been a drug target for chemotherapy of cardiovascular diseases at present. The objective of our study is to synthesize and study bifunctional or multifunctional enzyme inhibitors in mevalonate pathway. Based on the preliminary studies by our group, we intended to replace the P-O bond in the former inhibitors by P-C bond ans P-N bond that may be more resistant to enzymatic cleavage, and less likely to be rapidly dephosphorylated under normal conditions by several enzymes. A series of substrate analogues were synthesized in the present study and their activities were assayed. The preliminary results showed some of them were good inhibitors for the enzymes in mevalonate pathway and may be potentially better drugs. |
