Analysis of scopoletin and rutin in Noni (Morinda citrifolia) juice by LC-TOF/MS and HPLC

Scopoletin and rutin in Noni (Morinda citrifolia) juice from Cook Islands were identified by LC-TOF/MS method and quantified by RP-HPLC method with UV detection. The two compounds were identified on TOF/MS by comparing the response with that of their standard. The result proved the existence of scopoletin and rutin in Noni juice. The HPLC assay was performed under the following conditions: a reversed-phase Alltima C18 column (250×4.6 mm, 5µ), methanol and 0.01% formic acid solution (1:1, V/V) as the mobile phase, sampling rate of 0.7 mL/min, photodiode array detector (PAD) as the detector. An external standard method was used for the accurate quantification of two compounds. The results show that the developed method for the determination of scopoletin and rutin in Noni juice has a good precision with the RSD values of 0.68% and 0.85% respectively, and good recoveries with the RSD values of 1.82% and 1.76% respectively. The contents of scopoletin and rutin in Noni juice were 0.075 mg/mL and 0.017 mg/mL respectively.
