In this paper, a method was developed for the determination of the contents of polysaccharides in Semen Plantaginis. The contents of polysaccharides in Semen Plantaginis were determined by the Phenol-H2SO4 colorimetry method at 480 nm. In this method xylose was chosen as the standard for the determination. The results show that the average value of content of polysaccharides in Semen Plantaginis is 9.04%, the mean recovery rate for the determination of the polysaccharides is 98.2% with the RSD of 1.71% (n=6). The developed method is simple, accurate and reproducible for the determination of polysaccharides in Semen Plantaginis. It could also be suitable for the determination of polysaccharides from other plants in which there are high contents of pentoses.
