Molecular cloning and characterization of a new sesquiterpene synthase in Streptomyces avermitilis

The Streptomyces avermitilis genome contains one gene that shows high levels of similarity to the gene for epi-isozizaene synthase previously isolated from Streptomyces coelicolor. This gene has been amplified by PCR, cloned into a pET vector and expressed in E. coli. After purification of the inclusion body with an ion - exchange column, the recombinant protein has been characterized. It catalyzes the cyclization of farnesyl diphosphate to epi-isozizaene, a sesquiterpene hydrocarbon product which has rarely been observed as a natural product.
