| A rapid, sensitive and specific method to quantify Olanzapine in human plasma using Mirtazapine as the Internal standard (IS) is described. The analyte and IS were extracted from plasma by liquid-liquid extraction using tertiary butyl methyl ether (TMBE) solvent. After removal and dried of organic phase the extracts were reconstituted with a fixed volume of mobile phase (20:80:10M Ammonium formate: ACN). The extracts were analyzed by LCMS/MS. Chromatography was performed isocratically on X-terra RP C18 50°Á4.6 mm 5µ column, the method had a chromatographic run time of 2.5 min. and a linear calibration curve over the range 0.104ng /mL ¨C 23.930ng/mL (r2=0.9998). The limit of quantification was 0.104ng/mL that indicates the method have high sensitivity. Experimental design was used for validation of robustness as instrumental factors such as flow rate, % acetonitrile in mobile phase, auto sampler temperature and column over temperature, this experimental design also applied for sample preparation factors such as TBME volume, auto pulsar time and solvent evaporation and antioxidant concentration considerable influence was observed with flow rate and TBME volume. The intra and interday precision was less than 0.82. number of interferences were found from endogenous compounds. The method was suitable for pharmacokinetic and bioequivalence studies of Olanzapine. In this presentation I will explain the importance of experimental design for robustness. |
