Rapid on-chip immunoassays for detection of endogenous disease biomarkers in saliva

Diagnostic technologies capable of providing rapid, reliable assessment of local and systemic health, especially for early diagnosis of disease, would significantly positively impact clinical medicine. The functionality required for such diagnostics demands compact instrumentation having analytical performance approaching that of conventional laboratory assays. We have developed a microfluidic-based clinical diagnostic that enables rapid quantitation of identified molecular disease biomarkers in human saliva using on-chip homogeneous electrophoretic immunoassays. We report on measurements of collagen-cleaving enzymes implicated in periodontal disease in saliva samples from healthy and periodontitis-afflicted subjects. Concentrations of endogenous enzyme can be measured in less than ten minutes over the clinically-relevant range of 100-5500 ng/mL. The dynamic range of the assay can be readily optimized by varying the duration of sample enrichment. The assay is fast, requires as little as ten microliters of saliva, and is pertinent to quantitation of disease markers in several body fluids.
