The RIT Proteomics lab has been using 2D-gel electrophoresis analysis of proteins to study different known human carcinogens and the effects of these compounds on protein expression in the bacteria Pseudomonas putida. The compound being studied is diethylstilbestrol (DES). After proper separation of the proteins using the 2D-gel electrophoresis, the protein expression for the bacteria in the presence of the carcinogen DES can be seen. Protein expression levels can be analyzed using Phoretix 2D software which detects changes in the protein levels, when the bacteria are grown in the presence and absence of DES. Protein spots of interest can be digested with trypsin and then analyzed by mass spectrometry to identify the peptide sequence of the protein. |
