| Multidimensional protein identification technology is a powerful approach for determining the content of multiprotein complexes. We are implementing novel ways of visualizing protein complexes and protein interaction networks that take into account the abundance of a protein present in a MudPIT analysis after an affinity purification scheme. By implementing a straightforward approach named the normalized spectral abundance factor, replicate analyses of samples are readily compared and the abundance of proteins in complexes can be determined. This seminar will demonstrate the use of this approach for visualizing distinct forms of the mammalian mediator complex, a transcriptional regulatory complex, and for probabilistic assembly of a protein interaction network. |
