| The goal of this research is to characterize by gas chromatography and mass spectral analysis the oxidation products of arachidonic acid and eicosapentaenoic acid metabolism by the enzyme prostaglandin H2 synthase, better known as cyclo-oxygenase.This enzyme comprises two catalytically distinct reactions involving cyclization of arachidonic acid by the cyclooxygenase activity, followed by the peroxidase activity. The reagent trimethyl silyl diazomethane(TMSD) has been successfully used in our laboratory to esterify polyunsaturated fatty acids and is currently being investigated as a reagent for esterification of their oxidation products, some of which are prostaglandins. GC-MS analysis for identification and quatification of prostaglandins is accomplished following their chemical modification to render them more readily volatile and/or stable.
