Purification of cellobiohydrolase from Fomitopsis pinicola for the enzymatic saccharification of lignocellulosic biomass

The brown rot funus Fomitopsis pinicola was able to hydrolyze lignocellulosic biomass. The purification of cellobiohydrolase related to degradation of crystalline cellulose of lignocellulosic biomass was carried out. By the hydrolysis using F. pinicola on various carbon sources, the total sugar yield on pretreated wood powder was 0.85 mg/g (biomass dry weigh). For the analysis of related enzyme on hydrolysis, the activities of extracellular enzymes of F. pinicola were determined on carbon source as avicel, wood powder (Pinus densiflora) and pretreated wood powder. The highest activity of cellobiohydrolase was obtained at shaking culture containing avicel as carbon source. For the characterization and purification of cellobiohydrolase which hydrolyzed avicel as crystalline cellulose, culture solution was purified by FPLC. According to the result of SDS-PAGE of purified enzyme, protein band was observed at about 70 kDa. The optimum pH and temperature was 5.0 and 60°C for cellobiohydrolase activity, respectively.
