| Three highly reactive non-chromophoric reagents (trifluoromethanesulfonic anhydride, trifluoroacetic anhydride, and methanesulfonic anhydride) are widely used in organic syntheses. These reagents pose challenges to potency measurements due to their sensitive nature in regards to water, alcohols, and HPLC column surfaces. A chromatographic potency method has not been reported in the literature. By derivatizing the anhydrides with benzylamine, potency determinations are possible using HPLC because the resulting amides are not only stable but also detectable by UV. The specific derivatization conditions and procedure to minimize side reactions and obtain acceptable accuracy is presented. Linearity and precision were performed and the method was proven to be accurate and precise. |
